Postdoc Controlling tissue formation in situ cardiovascular tissue engineering
The position: Eindhoven University of Technology (TU/e) has a postdoctoral research position available in the group Soft Tissue Biomechanics and Engineering within the Department of Biomedical Engineering. The group has a strong interdisciplinary character, bridging the fields of regenerative medicine, biomechanics, mechanobiology, immunomodulation, and biomaterials science, and applies this to develop novel technologies for in situ tissue engineering for cardiovascular applications. The group has excellent research facilities including a certified cell culture lab for working with human and animal cells and tissues, analytical facilities for microscopy and mechanical testing and conditioning of cells and tissues, and advanced scaffold construction equipment. Moreover, advanced computational platforms are available for developing computational frameworks aimed at the prediction and understanding of cardiovascular regeneration. This post-doctoral position is part of a project granted by the Netherlands CardioVascular Research Initiative (CVON-1Valve).
The project: The overall aim of the CVON-1Valve project is to employ in situ tissue engineering to create a readily-available, living heart valve replacement that can grow with the patient and adapt to functional demand changes without the need for reoperations: one valve for life. This would overcome the limitations that are associated with currently available heart valve prostheses and thus answer to the persistent clinical need for improved heart valve replacements. The in situ tissue engineering strategy relies on the use of “smart” bioresorbable synthetic heart valve scaffolds, which accommodate host cell infiltration and stimulate the formation of new tissue, directly in the functional implantation site. The main challenges lie in understanding and controlling the formation of functional new tissue by endogenous cells in the stringent hemodynamic environment.
CVON-1Valve is a collaborative research project between Eindhoven University of Technology, Erasmus University Medical Center, Utrecht University Medical Center, Academic Medical Center Amsterdam, the University of Zürich, and the Dutch Heart Foundation, and combines basic research on in vitro and in vivo models with long-term pre-clinical studies and early Health Technology Assessment.
The role: The TU/e postdoctoral researcher will be responsible for:
- The development of a predictive, combined in vitro/in silico model to enable the rational development of new scaffolds for in situ cardiovascular tissue engineering.
- The establishment of a dynamic 3D in vitro (co-)culture model, based on existing models in our facility, to study cell-scaffold interactions in a physiologically relevant biomechanical environment.
- Application of the in vitro model to correlate the initial immunomodulatory effects of 3D scaffolds with the downstream formation of new tissue.
- Development of theoretical and computational frameworks to predict and understand early tissue formation as observed experimentally with the in vitro platform.
- Utilize computational models to extrapolate in vitro findings to the more complex in vivo conditions, to predict and understand the long-term pre-clinical results.
The Person: The ideal candidate holds a PhD in either bio(medical) engineering or mechanical engineering or has a strong background in a related field. Experience with cell/tissue engineering techniques and immunomodulatory biomaterials is considered an advantage, as well as experience with computational modeling and continuum mechanics. More importantly, the candidate is curious, flexible, independent and highly motivated. The candidate should have excellent communication skills that support the work in a multidisciplinary project team.
